Project Title: Alteredcell-cell interactions in experimental diabetic neuropathy - translation of mouse to human

Source of funding: DFG GRK1874-1 DIAMICOM (18 month with the perspective of prolongation to 36 month)

Project description: Several studies which have investigated the underlying pathophysiological mechanisms of diabetic neuropathy (DN) have relied on experiential models, in particularly the treatment with streptozotocin (STZ). However, recent data suggest that STZ has neurotoxic effect. It is therefore imperative to translate concepts observed in STZ diabetes to the human situation. Utilizing a newly established biobank of peripheral nerves from individuals with and without diabetes, this project will seek to determine the morphological and biochemical changes that occur in peripheral nerve experimental and clinical DN and whether these changes are caused by in the loss of defence mechanisms against reactive metabolites.

Methods that will be used:
  • Analyze of peripheral nerves and dorsal root ganglia (DRG) from healthy and diabetic C57Bl/6 wild-type, Glo1-/- mice
  • Study of the onset and progress of neuronal dysfunction by function assays (mechanical and thermal nociception using Hot-plate, Hargreaves, Tail-flick and Frey-filament assays), determination of motor and sensory nerve conduction velocity (NCV) and neuronal blood flow (NBF)
  • Histology and flow cytometer analysis to determine changes in morphology and the various cell populations within the nerve, particularly in relation to infiltrating cells
  • Biochemical analysis of the key enzymes involved in the detoxification of reactive metabolites, such as the glyoxalase system, aldo-keto reductase and MnSOD. Furthermore, the post-translational modifications associated with reactive metabolites, such as methionine sulphoxide, dityrosine and methylglyoxal-derived hydroimidazlone (MG-H1), would be determined by histology, western blotting and liquid chromatography, or tandem mass spectroscopy
Profile of candidate’s qualification: We are looking for a highly motivated PhD student (m/f) interested in diabetic microvascular research and regenerative medicine, to perform competitive high quality scientific research in a stimulating environment. She or he will have a Masters Degree in Life Sciences acceptable to HBIGS and a strong background in molecular biology and/or cell biology. Previous experience in working with cell culture and molecular/cell biology techniques would be an advantage.
